Background
Natural products have historically been the most important source of drugs, with medicinal mushrooms being used in traditional medicine for their immune-modulating and anti-cancer properties. Cyathus striatus (CS) is a higher Basidiomycetes mushroom that produces bioactive compounds including indole substances, diterpenoid antibiotics, and cyathins. Previous research found that ethyl acetate extracts from CS induced apoptosis in pancreatic cancer cells through multiple pathways.
Objective
To identify and characterize the structure of active compounds in Cyathus striatus that demonstrated anti-pancreatic cancer activity in previous studies, and to evaluate their biological effects on human pancreatic cancer cells.
Results
The main active compound was identified as striatal C with molecular formula C25H34O7. The purified striatal C significantly reduced pancreatic cancer cell viability in a dose-dependent manner, with maximal effects after 8 hours of treatment. At 10 μg/mL, striatal C inhibited growth by 85% and 75% in HPAF-II and PL45 cells respectively. The compound induced apoptosis as evidenced by increased sub-G1 phase cells, positive Annexin-V staining, and DNA fragmentation in TUNEL assays.
Conclusion
This study identified striatal C as the main active compound in Cyathus striatus extract responsible for inducing apoptosis in human pancreatic cancer cells. The findings suggest striatal C has potential as an anti-cancer therapeutic agent, though further research is needed to develop synthesis methods and conduct clinical studies.
