β-1,6-Glucan from Pleurotus eryngii Modulates the Immunity and Gut Microbiota

Background

Pleurotus eryngii (king trumpet mushroom) has been widely used as an edible mushroom with various health benefits including anti-inflammatory, antioxidant, antitumor and antibacterial properties. These effects are attributed to active ingredients like polysaccharides, peptides, and sterols. β-1,6-glucan is an important component of fungal cell walls that has shown immunomodulatory, anti-tumor and anti-oxidant activities.

Objective

To obtain a homogeneous branched β-1,6-glucan (APEP-A-b) from P. eryngii fruiting bodies and investigate its effects on immunity and gut microbiota in mice.

Results

APEP-A-b significantly increased splenic lymphocyte proliferation, NK cell activity and phagocytic capacity of peritoneal cavity phagocytes. The proportion of CD4+ and CD8+ T cells in lamina propria increased significantly. Treatment led to increased abundance of beneficial bacteria like Lachnospiraceae and Rikenellaceae families, while reducing pathogenic bacteria. APEP-A-b also increased concentrations of acetic and butyric acid in cecum contents.

Conclusion

The β-1,6-glucan isolated from P. eryngii enhances immunity by modulating gut microbiota composition and metabolites. The 200 mg/kg·d dose showed optimal immunomodulatory effects. These findings provide insights for developing β-1,6-glucan as a therapeutic agent to enhance immunity.
