Prospective Comparative Study of an Oral Synbiotic and a Myoinositol-Based Herbal Supplement in Modifying Hormone Levels and the Gut Microbiome in Non-cystic Acne

Summary
This study tested two supplements for treating non-cystic acne: one containing probiotics and herbs, and another based on myoinositol and medicinal plants. Both supplements significantly reduced acne lesions and improved gut health by increasing beneficial bacteria and beneficial fatty acids produced by these bacteria. The myoinositol-based supplement also reduced hormone precursors related to acne development. These findings suggest that supporting gut health may be an effective way to treat acne.
Background
Acne pathogenesis involves multiple systemic factors including gut dysbiosis, hormonal imbalances, and chronic inflammation. Probiotics, myoinositol, and plant-derived molecules have shown potential in modulating acne through these pathways.
Objective
To compare the efficacy of a synbiotic supplement containing herbs versus a myoinositol-based herbal supplement in improving non-cystic acne and to evaluate their effects on gut microbiome composition, short chain fatty acids (SCFAs), and hormonal profiles.
Results
Both supplements significantly reduced acne lesion counts at weeks 4 and 8. Several beneficial bacteria increased threefold or more in both groups, including Faecalibacterium prausnitzii and Prevotella copri. Stool and plasma SCFAs increased significantly in both groups. The myoinositol-based supplement significantly reduced 17-OHP and androstenedione after 8 weeks.
Conclusion
Both supplements improved non-cystic acne through multiple mechanisms including enhanced gut microbiota composition, increased SCFA production, and hormonal modulation. The myoinositol-based supplement additionally reduced androgen precursors, suggesting a specific hormonal mechanism of action for acne reduction.
